If all three materials are present but the craft is greyed out, the likely blocker is Mastery or the recipe technology.
Ash comes from cremating a corpse.

So burning a poor corpse does not mean giving up the normal certificate income.
A basic cremation gives enough Ash for multiple Ink batches.
That makes low-value corpses useful even when they are bad grave candidates and not worth turning into zombies.

The Crematorium is part of morgue/corpse progression.
Once the appropriate technology is available, build it in its designated area.
If you are cremating your first corpse specifically for Ink, avoid spending all of the Ash on random experiments before the writing quest is solved.
Ash has many uses, but early writing progression is a common immediate need.
Water is the easiest ingredient.
Draw it from your well.
Later upgrades make water production more convenient, but the first Ink batch does not require any elaborate setup.
The Writing Desk needs the Water resource itself, not an Empty Bottle or some other container.
Melted Fat is produced in the Distillation Cube.

For a body intended for burial, that is a particularly efficient choice: Fat contributes a red skull, so removing it both improves the corpse and supplies Ink-related production.

A quest asking for a little paper and Ink therefore quietly makes sure you have opened a much larger piece of the game's infrastructure.
You did not miss a simple village ink bottle early on. Before Town access, crafting is the intended route.

So one batch is enough for the Ink requirement and leaves two spare.
Do not automatically make several batches while Melted Fat is still a scarce resource.

The Writing Desk can exist before you comfortably meet every recipe's Mastery requirement.
You need exactly Ash, Water and Melted Fat.
If you rely on shared storage, make sure the Writing Desk can actually see the required materials in its area.
